Abstract:
using (C_(57) ×cba) F_1 mouse sperm and NIH mouse in vitro fertilization, the chromosome samples of fertilized eggs were prepared by culture, and the Methyl methanesulfonate (MMS) was observed. Effect on chromosome aberration of mouse sperm. MMS in vivo (25,50, 150 mg/kg), in vitro (75, μg/ml) after the treatment of sperm, sperm chromosome aberration rate increased significantly, dose-response relationship. The main type of distortion was staining type. This study shows that sperm DNA damage induced by MMS can be transmitted to spermatozoa and eggs; The model used is a reliable method to monitor the mutagenic effect of chemicals on germ cells (sperm, eggs).

Abstract:
objective: to investigate the relationship between telomerase activity and growth status of CAOV3 cells treated with Methyl methanesulfonate (MMS). Methods: trypan blue staining method to count live cells, thiazole Blue (MTT) method to determine the drug concentration, flow cytometry to observe cell apoptosis, telomeric repeat amplification assay (TRAP), telomerase activity was detected by polyacrylamide gel electrophoresis and silver staining. Results: there was no significant difference in the percentage of viable cells between the treated group and the untreated group. MMS mainly blocked CAOV3 cells in S phase, and no apoptotic peak was observed. Compared with the control group, the telomerase activity was increased in the treatment group. Conclusion: MMS can up-regulate telomerase activity in CAOV3 cells. The anti-apoptotic effect of CAOV3 cells on MMS may be related to the up-regulation of telomerase activity.
