| In vitro study | WY 14643 (10 μm) acts on aortic smooth muscle cells and almost completely inhibits IL-1 of the induced production of prostaglandins and cyclooxygenase-2 by inhibiting the NF-κB signaling pathway. WY14643 (250 μm) significantly reduced the expression level of TNF-α stimulated human endothelial cells by VCAM-1 and by 52%. Pretreatment of endothelial cells with WY 14643 (10 μm) followed by stimulation with TNF-α reduced U937 cell adhesion by 50%. |
| In vivo study | WY 14643 (1 mg/kg IV bolus) treatment of rats for 30 minutes resulted in left anterior descending occlusion after regional myocardial ischemia (25 minutes) and reperfusion (2 hours), infarct size was significantly reduced by ~ 44%. WY 14643(3 mg/kg) treatment of high-fat-fed rats reduced plasma glucose, triglycerides (-16% vs untreated group),Leptin(-52%), muscle triglyceride (-34%), total long chain acyl-CoA (LCACoAs)(-41%) levels. WY14643 significantly reduced visceral fat weight and total hepatic triglyceride content without increasing body weight. WY14643 enhances the sensitivity of the whole body to insulin. WY 14643 enhances the insulin-mediated index of muscle glucose metabolism (Rg') in red (47%) and White (63%) muscle, and in white adipose tissue (90%), and reduces the accumulation of triglycerides and lcacoas in muscle. |

WY-14643 is a peroxisome proliferator-activated receptor alpha (PPAR) agonist. Therefore, WY-14643 is used in the study of lipid metabolism, liver diseases and obesity.
Preparation Method:
