Abstract:
The application of two different deproteinization methods (enzyme Sevage method and trichloroacetic acid method) in the extraction process of polysaccharide from Spirulina platensis was compared. The results showed that different methods of removing protein not only affected the removal rate of protein and the yield rate of polysaccharide, but also the composition, molecular size and monosaccharide composition of spirulina polysaccharide were different.

Abstract:
The anticancer effect of polysaccharide from Spirulina platensis was studied in vitro and in vivo. 0. The inhibition rate of 3~1.5g · L-1 on B3 7 breast cancer cells was up to 8.0%, and the inhibition rate on K562 leukemia cells was 46. The IC50 of the two cells were 1.4 8 and 1.56g · L-1, respectively. When polysaccharide from Spirulina platensis was administrated by gavage at 30 150 and 75mg · kg-1, the inhibition rates of S180 sarcoma in mice were 55.2%, 4 4. The inhibition rates of H2 2 mice were 33.8% and 7.1%, respectively. 4% and 31.0%; The tumor inhibition rate of EAC mice was 56 .9%, 4/4. 7% and 2. At the same time, the spleen index and thymus index of tumor-bearing mice were significantly increased. MTT method was used to detect the transformation of spleen lymphocytes and the effect on NK cell activity of spirulina polysaccharide in tumor-bearing mice. It also showed that it could improve the lymphocyte transformation rate and promote NK cell killing target cells. Determination of spirulina polysaccharide on spleen lymphocyte TNF-α induced effect, high dose group supernatant reached 75 × 10 -3 IU · L-1 (control group 34 × 10 -3 IU · L-1), and a better dose-response relationship. After PSP treatment, the function of spleen lymphocytes in tumor-bearing mice was enhanced to produce interleukin-2 (IL-2). Abstract:
The traditional process of extracting polysaccharide from Spirulina platensis has the problems of long process, complex operation and large consumption of organic solvent. Trichloroacetic acid (TCA) method was used to remove protein instead of Sevag method, which increased the protein removal rate, reduced the Polysaccharide loss rate and shortened the process in the improved extraction process. Two polysaccharide fractions were obtained by separation and purification with DEAE-Speharose and Sephadex-G50 columns. The relative molecular mass was Mr1 = 12 600 and Mr2 = 16 600, respectively. Monosaccharide composition of component 1: D-glucose, D-mannose, D-galactose and glucuronic acid; Monosaccharide composition of component 2: D-glucose, D-xylose, L-rhamnose and glucuronic acid.
