In vitro studyIrreversible inhibition of Tideglusib by GSK-3 in human neuroblastoma cells and murine primary neurons reduces tau protein phosphorylation levels and prevents apoptosis. Tideglusib (2.5 μm) inhibits glutamate-induced activation of glial cells, evidence for this conclusion is a decrease in the expression levels of TNF-α and COX-2 in rat primary astrocytes or microglia. Tideglusib (2.5 μm) also has a potent protective effect on cortical neurons from glutamate-induced excitotoxicity, evidence for this conclusion is a significant reduction in the number of Annexin-V positive cells in rat primary astrocytes or microglia

In vivo studyTideglusib (50 mg/kg) injection into the hippocampus of adult male Wistar rats significantly reduced kainic acid-induced inflammatory responses and had a protective effect on damaged areas of the hippocampus. In APP/tau double transgenic mice, Tideglusib (200 mg/kg, P. O.) reduced tau phosphorylation, reduced amyloid deposition and plaque-associated astrocyte proliferation, protection of neurons in the entorhinal cortex and hippocampal CA1 region from death and prevention of memory loss
Tideglusib (50 mg/kg) injection into the hippocampus of adult male Wistar rats can greatly reduce the inflammatory response induced by kainic acid and has a protective effect on the damaged area of the hippocampus. In APP/tau double transgenic mice, Tideglusib (200 mg/kg, P. O.) reduced tau phosphorylation, reduced amyloid deposition and plaque-associated astrocyte proliferation, protection of neurons in the entorhinal cortex and hippocampal CA1 region to prevent death and memory loss

Tideglusib - Introduction

Tideglusib is a small organic molecule compound with the chemical name of N-(6-bromo-2-pyridyl)-2,6-difluoroaniline guanidino methanesulfonamide. It is a white or off-white crystalline solid, stable at room temperature. The molecular formula of Tideglusib is C19H14BrF2N3O3S, and the relative molecular mass is 491.3.

The primary use of Tideglusib is as a protein kinase C(PKC) inhibitor. PKC is a class of enzymes involved in biological processes such as signal transduction, cell proliferation and apoptosis. The inhibitory effect of Tideglusib can regulate the activity level of PKC, which has potential therapeutic effects on some neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease and Huntington's disease.

The method for preparing Tideglusib includes the reaction of N-(6-bromo-2-pyridyl) -2,6-difluoroaniline and ethyl methanesulfonate, it is then converted to the target product of Tideglusib by a series of chemical reactions. The preparation process requires control of temperature and reaction conditions to ensure high purity and yield of the product.

Regarding the safety information of Tideglusib, according to existing research, it is usually relatively safe at recommend doses. However, for any chemical substance, the relevant safety data sheet and instructions should be carefully read before use, and the instructions for use should be strictly followed. When using or handling Tideglusib, care should be taken to avoid contact with skin and eyes and to ensure good ventilation conditions. If any symptoms of discomfort or intoxication appear, seek medical assistance immediately. In addition, because Tideglusib is a laboratory-grade chemical, its clinical safety and effectiveness still need further research and verification.
