| In vitro study | Pregnenolone dose-related induction of MAP2 induces the rate and extent of tubulin assembly, while progesterone, inactive by itself, counteracts the stimulatory effect of Pregnenolone. Pregnenolone induced an increase in microtubule assembly, resulting in a completely normal structure. Pregnenolone maintains microtubule abundance and promotes cell movement during the outsourcing process. In the mouse hippocampal cell line (HT-22), Pregnenolone caused a significant decrease in the localization of the GR nucleus. Pregnenolone has neuropathological neuroprotective effects against glutamate and beta amyloid. |
| In vivo study | In intact male SCID mice and castrated animals, Pregnenolone maintained its in vivo proliferative activity and stimulated the growth of LNCaP tumor xenografts. Pregnenolone activates transcription via the LNCaP androgen receptor (AR), but not wild-type AR. In intact male SCID mice and castrated animals, Pregnenolone caused a significant decrease in astrocytes in the vicinity of the wound and caused a significant decrease in bromodeoxyuridine incorporation in rat active astrocytes. Pregnenolone leads to an increase in downstream neurosteroids such as allopregnanolone, which has a neuroprotective effect, increases neurogenesis, reduces apoptosis and inflammation, regulates the hypothalamic-pituitary-adrenal axis, and significantly increased the GABA(A) receptor response. Pregnenolone raises Pregnenolone sulfate, which is a neurosteroid that actively regulates NMDA receptors. |
