| In vitro study | Milnacipran is excreted mainly in the form of bulk and glucuronide through urine (>80%), and only a small part (<10%) through the N-terminal deethylation catalyzed by CYP3A4, Milnacipran can inhibit certain ligand ion channel (LGIC) receptors, including NMDA, 5-HT3A and nACh receptors, at high concentrations, the IC50 values were 58.4 μm, 185 μm, and 14.3 μm, respectively. |
| In vivo study | Milnacipran(10 and 30 mg/kg, P. O.) increased extracellular 5-HT and NA levels in the rat prefrontal cortex in a dose-dependent manner. In an animal behavioral model of depression, Milnacipran (30 and 60 mg/kg, P. O.) significantly reduced resting time in a compulsive swimming experiment in rats. In an animal behavioral model of anxiety, Milnacipran (30 and 60 mg/kg, P. O.) also significantly reduced immobility time in the conditioned fear stress test in rats. Milnacipran (<40 mg/kg I. P.) increased extracellular NA and 5-HT levels in the hypothalamus of freely moving guinea pigs in a dose-dependent manner. In the hypothalamus of freely moving guinea pigs, treatment with 10 mg/kg and 40 mg/kg Milnacipran, respectively, reduced the nap metabolite MHPG by 57% and 47%, respectively. |
