activity Study | Wang Ming et al. discussed a new Arsenic Iodide complex Arsenic Iodide-thiourea (AsI3-Tu) effects on proliferation and survivin gene expression of leukemia HL-60 cells. Methods: HL-60 cells were treated with different concentrations of arsenic iodide-thiourea, cell proliferation activity was detected by MTT colorimetric method, cell apoptosis was observed by in situ end labeling (TUNEL) method, RT-PCR semi quantitative detection of sur-vivin mRNA expression in cells. Results: ①AsI3-Tu of each experimental group could inhibit the proliferation of HL-60 cells, which was related to the time and dose. (2) TUNEL method was used to observe the apoptosis, and the number of apoptotic cells in AsI3-Tu concentration groups was significantly increased. ③The expression level of survivin mRNA was decreased in AsI3-Tu groups, and the expression level was negatively correlated with the dose of AsI3-Tu. Conclusion: AsI3-Tu can effectively inhibit the proliferation of leukemia cell line HL-60 and induce its apoptosis. The mechanism may be related to the inhibition of survivin gene expression in HL-60 cells. |
