HpaII - Introduction
It is a restriction enzyme, which belongs to DNA methylation sensitive enzyme. It can recognize the CpG methylation site of the target DNA sequence and cleave the specific CpG methylation site in the mixed enzymatic hydrolysis reaction.
The use of
has two main aspects. First, it can be used for methylation-specific PCR(MSP) or restriction fragment length polymorphism analysis (RFLP) to study the methylation status of DNA and changes in specific methylation sites. Secondly, it can also be used for the identification of methylated DNA sequences and the digestion and purification of methylated DNA fragments.
The preparation method of
is to transform Escherichia coli containing the gene, and then use the enzyme secreted by it for purification. The purification process usually includes the steps of cell culture, membrane protein extraction and enzyme activity detection.
Regarding safety information, enzyme reagents are commonly used in biological laboratories and are generally considered to be relatively safe. However, any enzyme reagent should be careful to follow proper laboratory practices and safe handling measures. When using, should wear gloves, avoid direct contact with skin and mucous membrane. In addition, the relevant waste disposal regulations should be followed to dispose of the waste correctly. In the event of an accident, appropriate emergency treatment and medical treatment should be carried out immediately.
