Introduction | (R)-N-tert-butoxycarbonyl-3-iodoalanine methyl ester is an organic intermediate, which can be obtained by reacting with elemental iodine Boc-L-serine methyl ester as raw material. |
preparation | Boc-L-serine methyl ester (20g,91mmol) is dissolved in THF(400mL) and cooled in an ice water bath. Triphenylphosphine (36g,137mmol) and pyridine (14g,177mmol) were added to the upper solution in turn, and then I2(34g,134mmol) was added in batches to maintain the temperature below 20 ℃ for 4 hours. After the reaction is completed, add ice water to the reaction solution to quench the reaction and extract it with ether. The organic phase obtained was washed with 1% hydrochloric acid, 5% Na2SO3 solution and saturated NaCl solution, dried with anhydrous Na2SO4, and concentrated after filtration. The resulting residue is added to petroleum ether, stirred to precipitate solids, filtered, and the filter residue is washed with petroleum ether. The filtrate was concentrated to get 26g of white or yellowish solid with 87% yield. |
