In vitro studyAmikacin sulfate has high affinity for 16S rRNA of bacterial 30S ribosome, causing mRNA mistranslation and making bacteria unable to synthesize proteins important for its growth. Amikacin in vitro against Pseudomonas spp., Escherichia Coli, Proteus (indole-positive and indole-negative), Providence spp., Klebsiella-Enterobacter-Serratia spp, acinetobacter spp. (original name Mima-Herellea), and Citrobacter freundii are active. In vitro studies have shown that amikacin sulfate in combination with β-lactam antibiotics acts synergistically on a number of clinically important Gram-negative organisms. In vitro studies have shown that amikacin sulfate in combination with β-lactam antibacterial agents, including moxalactam or piperacillin, acts synergistically on a number of clinically significant Gram-negative organisms.
In vivo studyIn an experimental rabbit model of endocarditis, Amikacin (80 mg/kg I. v.) significantly reduced aortic valve neoplasm Pseudomonas density compared with controls. In addition, once-daily administration of amikacin was more effective than twice-daily administration. Amikacin, administered once, twice, and three times separately at a dose of 50 mg/kg, showed significant activity in a model of avian Mycobacterium tuberculosis complex infection. The addition of Clofazimine significantly increased the activity.
