In the current prior art, the preparation method of the non-natural amino acid N-fluorene methoxycarbonyl-β-(2-thienyl)-D-alanine and its intermediates is as follows: 2-thiophene methanol is acylated into 2-chloromethylthiophene by hydroxy chloride, and then mixed with sodium metal and ethanol and diethyl acetaminomalonate to synthesize diethyl ester, and then added D-acylase, the fmoc-d-thi-oh is prepared by reacting with fmoc-group. The operation process of this method seems simple, but it requires relatively expensive D-acylase, and the cost is high, so it is not suitable for commercial production.
