| Overview | Lupeol is a five-ring three-anvil compound that can be extracted from various fruits and vegetables such as strawberries, mangoes, and Chinese herbal medicines. It has anti-inflammatory, Anti-oxidation, promotion of wound healing and other pharmacological effects, and shows anti-cancer activity in pancreatic cancer, breast cancer, prostate cancer, melanoma and other tumors, it is clinically used for the treatment of leukemia, liver cancer, colon cancer, pancreatic cancer and other diseases. |
| pharmacological action | 1. anti-tumor effect (1) inhibitory effect on melanoma lupeol can inhibit the growth and induce differentiation of mouse melanoma. Induction of melanoma cell differentiation is achieved by activating the mitogen-activated protein kinase (MAKP) pathway, and the similarities and differences in the structure of the plume fan ring C- 3 play a key role in the activation of the MAKP pathway. Lupeol inhibits the metastasis of malignant melanoma, and its mechanism is mainly related to Wnt/β catenin (catenin) signaling pathway, but not to MAKP pathway. (2) Inhibitory effect on skin cancer in mice Lupeol has inhibitory effect on the occurrence of skin cancer and the expression of tumor markers. (3) Inhibitory effect on breast cancer Lupeol has inhibitory effect on the growth of breast cancer cells. Lupeol has inhibitory effect on estrogen receptor (ER) negative and positive breast cancer cells in a dose-and time-dependent manner. It indicates that the inhibitory effect of lupeol on breast cancer cells does not depend on ER, but it has certain correlation. Effect of lupeol on human epidermal growth factor receptor 2(HER2) in breast cancer cells. (4) Inhibition of prostate cancer cells and induction of apoptosis In endocrine-related tumors, besides breast cancer cells, lupeol also has inhibitory effect on prostate cancer. Lupeol can inhibit the growth and induce apoptosis of prostate cancer cell lines LNCaP and CWR22Rr1 in a dose-dependent manner. Its mechanism is mainly that lupeol activates fas receptor-mediated apoptosis pathway. In addition, lupeol also has a significant inhibitory effect on the growth of prostate cancer xenografts. (5) Inhibition and induction of apoptosis on pancreatic cancer Lupeol can inhibit the growth of pancreatic cancer, and inhibition of Ras pathway can induce apoptosis of pancreatic cancer cells.
