XbaI (X after BamHI, A means Aerogenes) is an endonuclease that belongs to the family of restriction endonucleases. It is produced by a type of bacteria called Xanthomonas bacteria.

The enzymatic activity of XbaI depends on the temperature in the presence of magnesium ions, and the optimum temperature is 37°C. It recognizes a specific DNA sequence, the 5 '-TCTAGA-3', and divides the DNA into two pieces for two positions of that sequence. The cleaved DNA ends typically result in sticky ends that have single-stranded DNA overhangs, making the sticky ends easy to attach to other DNA molecules.

XbaI has a wide range of applications in molecular biology research. It is commonly used in DNA sequencing, subcloning, plasmid construction and other experiments. Due to its specific cleavage sequence, it can be used for the identification and localization of specific DNA fragments.

The preparation of XbaI is usually through genetic engineering methods, the corresponding DNA sequence in the Xanthomonas bacteria genome is put into a suitable expression vector, and the expression and purification are carried out by host cells such as E. coli.

In safe operation, pay attention to follow the laboratory's biosafety procedures when using XbaI. XbaI is an enzyme that is not directly toxic, but still requires careful handling and proper storage.
