TCTU - Introduction

Polypeptide reagent TCTU(N,N,N',N'-tetramethyl-O-(6-chloro-1h-benzotriazol-1-yl) urea hydrochloride tetrafluoroborate) it is a commonly used activation reagent, which is commonly used in peptide synthesis and protein chemical modification in solid phase synthesis.

Nature:
TCTU exists in the form of colorless crystals or white powder, and is soluble in organic solvents such as acetonitrile, dimethyl sulfite and dimethylformamide.

Use:
TCTU can be used to synthesize polypeptide chains or to introduce various functional groups. It is widely used in solid phase synthesis in the introduction of dihydropyrrolyl (Dp), C- terminal acylation modification and isotope labeling reactions.

Method:
The preparation of TCTU usually uses a series of chemical reactions. The most common method is the reaction of N,N-dimethylurea together with 6-chloro-1H-benzotriazole and then with tetrafluoroboric acid to give TCTU.
