| In vitro study | Dapoxetine not only reduced the Kv4.3 peak current but also accelerated the current inactivation decay rate in a concentration-dependent manner. Dapoxetine reduces the duration of the past polarization pulse of Kv4.3 current, IC 50 is 5.3 μm. Dapoxetine also caused an accelerated inactivation of a large number of off-states. Dapoxetine produced a significant use-dependent block with delayed recovery from inactivation of the Kv4.3 current. Dapoxetine reduced the peak amplitude of the current at Kv1.5 and accelerated the decay rate of current inactivation in a concentration-dependent manner, with IC 50 at 11.6 μm. Dapoxetine reduces the tail current amplitude and slows down the inactivation process of Kv1.5, which leads to the phenomenon of tail penetration. Dapoxetine produced a use-dependent Kv1.5 block at 1 and 2Hz frequencies and slowed the recovery of time-course inactivation. Dapoxetine also appears to be a useful adjunct to morphine, lowering the analgesic threshold, although Dapoxetine itself has negligible analgesic activity. Dapoxetine is the D-enantiomer LY243917, which is 3.5 times more potent as a serotonin reuptake inhibitor than the L-enantiomer. |
