| Preparation | 3-methoxyresorcinol (12.0g,85.6mmol) was placed in a flame-dried round bottom flask and CH2Cl2(130.0mL) was added under argon atmosphere. The suspension was cooled at 0 ℃ and AlCl3(22.8g,171.0mmol) was added. Continue stirring until the suspension becomes a clear yellowish solution (usually 10-15 minutes). Benzoyl chloride (14.9mL,128.0mmol) was added dropwise within 5 minutes. The reaction solution is heated to ambient temperature within 2 hours, and then quenched by carefully pouring it into an ice/water mixture. The water layers were extracted three times with CH2Cl2, the organic layers were combined, washed with brine, dried with anhydrous Na2SO4, and concentrated in vacuum. The crude residue was purified by rapid chromatography to obtain yellow solid-like cortexine. |
