| preparation | dissolving 29.32g of acrylonitrile in 300ml THF (tetrahydrofuran), cooling to 0 ℃, adding 29.05g of hydrazine hydrate dropwise at 0 ℃, and adding dropwise for 20min. The system reacts at room temperature for 2h. At room temperature, 79g p-methoxybenzaldehyde was added dropwise for 20min and reacted at room temperature for 13h. The reaction is complete, the reaction is stopped and concentrated. The concentrated system is dissolved in 320ml n-butanol, 68.21g potassium tert-butanol is added, and the temperature is raised to 120 ℃ for 2h. Stop the reaction, drop to room temperature, pour into ice water, extract EA (ethyl acetate) (100ml * 5), wash the organic phase with 2N hydrochloric acid (100ml * 6), wash the aqueous phase with 50% NaOH aqueous solution until alkaline, extract the aqueous phase (100ml * 5) from EA, dry and concentrate to obtain 94.21g of 2-(4-methoxybenzyl)-2H-pyrazole -3-ylamine with a yield of 83.98%, brown solid. TLC information: raw material Rf = 0.80, product Rf = 0.35. Development agent: PE:EA (petroleum ether: ethyl acetate) = 1:1. |
