The diterpenoid compound 17-hydroxy rock Euphorbium B isolated from the root of Euphorbia chamaejasme significantly inhibited the growth of K562 cells and induced their apoptosis in vitro, and found that it has significant anti-inflammatory effects. In addition, studies have shown that it can induce apoptosis of breast cancer MDA-MB-231 cells by inhibiting PI3K /Akt signaling pathway. Results The linear relationship was good in the range of 4.070~32.560 μg/m L, r = 0.999 9, the average recovery rate was 98.02%, RS was 1.02%. Conclusion The method is specific, rapid and simple, and can be used as a method for the determination of content of Euphorbidione B in Euphorbia chamaedii, which can provide relevant basis for the formulation of quality standard of Euphorbia chamaedii. |

Compared with the blank group, 20-40 μmol-1 Euphorbidione B inhibited the mRNA expression of Cyclin D1 and e IF4E, and the mRN expression and protein level of Cyclin D1 in the transfected IF4E-siRNA group were also inhibited, while the cell cycle was blocked in the G1 phase (P <0 01,P <0.01). Therefore, rock eupholide B can inhibit the proliferation of human breast cancer cells and arrest the cell cycle in G1 phase, which may be caused by inhibiting the expression of e IF4E and thus downregulating the expression of Cyclin D1. |
