| Preparation | 11.9g[75.7mmol] dibutyl formamide (99%) was added to 10g[75.7mmol] 4-piperidinamide (97%) in a suspension of 50ml n-propyl acetate at 20°C for 5 minutes. After 5 minutes, at 20 ℃, 18.91g[158.9mmol] thionyl chloride was added. The addition takes 45 minutes, where the temperature remains constant at 20°C. After the addition, the mixture was stirred for another 18 hours at 20°C. The suspension was filtered and the filter residue was washed with n-propyl acetate. After drying, 8.55g of colorless solids remained. By quantitative NMR spectrum analysis, the 4-cyanopiperidine hydrochloride content is 95% (weight/weight). From this, the yield is calculated as the 73% of the theoretical value. |
