Preparation | The synthesis steps of Fmoc-L-3-chlorotyrosine are as follows: 3-chlorotyrosine (113.5g,526mmol) is added to a round bottom flask equipped with a mechanical stirrer and temperature probe. Dissolve 3-chlorotyrosine in a solution of dioxane: water = 2:1(500mL). Sodium carbonate (55.8g,526mmol) was added and the solution was cooled to 0°C in an ice bath. Fmoc-OSu(177.5g,526mmol) was dissolved in dioxane (300mL) and added to the solution at 0 ℃. The reaction was stirred at 0°C for 5 hours. Heat the reaction to 0°C. Room temperature overnight. Concentrate the reactants until a volume of 100mL is reached. Add 0.5M potassium bisulfate until pH is 3. Use pH test paper (0-14) placed directly in the mixture to determine pH. The pH was determined by comparison with the charts contained in the pH strip. The solution was extracted with ethyl acetate (3 × 500mL). Combine organic layers and wash with water (500mL) and salt water (500mL). The organic layer was then washed with sodium sulfate and concentrated to a constant 8 (under vacuum) to obtain Fmoc-L-3-chlorotyrosine, a light beige solid. (204g, 95% purity) |
