| preparation | at 0 ℃, slowly add ditert-butyl dicarbonate (10.2g, 1.2 equivalent) and triethylamine (19.0 ml) to a solution of (S)-2-amino-5-methoxy-5-oxopentanoate hydrochloride (7.70g, 1.0 equivalent) in dioxane (30 ml) and water (15 ml), 3.5 equivalent), stir the mixed solution at room temperature for 4 hours, then remove the solvent dioxane under reduced pressure, clean the reactants with ether, remove organic matter, acidify the water phase with hydrochloric acid (1M) to pH 4, extract the water phase with ethyl acetate, dry the combined organic phase with anhydrous magnesium sulfate, and finally remove the solvent under reduced pressure to obtain the target product Boc-L-glutamic acid -5-methyl ester. |
