preparation | sodium nitrite (14.00g,200mmol) is dissolved in 75ml DMF and 100ml of water, cooled to 0 degree, under nitrogen protection, 3N HCl(23ml,68.9mmol) is slowly added dropwise, and the reaction is completed for 10 minutes. At 0 degrees, 6-bromoindole (5.00g,25.5mmol) DMF(35ml) solution was slowly dropped into the reaction solution, and the solution was reacted overnight at room temperature. Extract with ethyl acetate for 3 times, combine organic phase, wash with water for 3 times, wash with saturated salt, dry with anhydrous sodium sulfate, concentrate, and purify with silica gel column to obtain 6-bromo-1H-indazole -3-formaldehyde with 83.6% yield. |
