Preparation | (a)3-methyl -4-iodobenzoic acid: 20g(0.132 mol) of 175ml of 3-methyl -4-aminobenzoic acid (20%) was introduced into a three-necked flask. At -10 ℃, 11.9g(0.172mol) of sodium nitrite solution in 50ml of water was added dropwise, and the mixture was stirred for 2 hours. The solution was added dropwise to 35g(0.211 mol) of potassium iodide 35.2g(0.185 mol) of copper iodide and 175ml of sulfuric acid (20%) through ampoules frozen at -5°C. Stir the mixture for eight hours, filter the reaction medium, dissolve the obtained solid in ethyl acetate, wash with water, then wash with sodium sulfite solution, dry with magnesium sulfate, and evaporate. 24.4g(70%) of 3-methyl-4-iodobenzoic acid with a melting point of 205-10°C were collected. (B) Methyl 4-iodo-3-methylbenzoate: 24.4g(0.093 mol) of 250ml of methanol of 3-methyl-4-iodobenzoic acid was introduced into the flask, and 2.5ml of concentrated sulfuric acid was added dropwise. The mixture is refluxed for twelve hours, the reaction medium is evaporated, the residue is dissolved with ethyl acetate and water, the organic phase is decanted, dried with magnesium sulfate, and evaporated. The residue was ground in methanol, filtered, and collected to 21.9g(85%) of the desired methyl 4-iodo-3-methylbenzoate, with a melting point of 58-9°C. |
