Overview | 5 '-nucleotidase (5'- Nucleotidase,5 '- NT) is a hydrolase with low substrate specificity and can act on a variety of nucleotides. It is composed of 2 identical subunits, each subunit has a molecular weight of 70kD, a hydrophobic group and an enzyme catalytic active center outside the cell membrane surface. Widely exists in the human liver and various tissues, located on the cell membrane, and mainly exists in the bile duct and sinus space membrane in the liver. |
clinical | 5 '-NT determination in serum is mainly used for diagnosis of hepatobiliary system diseases and differential diagnosis of bone diseases. The increase of 5 '-NT activity in serum is mainly seen in hepatobiliary system diseases, such as obstructive jaundice, primary and secondary liver cancer, hepatitis, etc., and its activity changes are usually consistent with the activity of ALP. In skeletal diseases, such as tumor metastasis, osteitis deformity, hyperparathyroidism, rickets, etc., ALP activity is usually increased, but 5 '-NT is normal. Therefore, the simultaneous determination of ALP and 5 '- NT is helpful for the differential diagnosis of hepatobiliary system and skeletal system diseases. |
Use | 5 '-nucleotide enzyme (5'-NT) is a hydrolase with low substrate specificity and can act on a variety of nucleotides. This enzyme is mainly derived from the liver and widely exists in human tissues, such as liver, gallbladder, intestine, brain, heart, pancreas, etc. Located on the cytoplasmic membrane, this enzyme mainly exists in the bile duct and sinus space membrane in the liver. The determination of 5 '-NT is mainly colorimetric, which is troublesome to operate and difficult to automate, but reagents are easily available and suitable for application in grass-roots units. |
