In vitro study | ABC294640 significantly altered the ceramide/S1P ratio in MDA-MB-231 cells, consistent with inhibition of SK activity. ABC294640 inhibits tumor cell proliferation with IC50 values ranging from approximately 6 -48 μm and impairs tumor cell migration with loss of microfilaments. In A- 498,PC-3, and MDA-MB-231 cells, ABC294640 induces nonapoptotic cell death, lysosomal morphological changes, autophagosome formation, and increased acid vesicles. ABC294640 reduces e2-stimulated ERE luciferase activity in MCF-7 and ER-transfected HEK293 cells. |
In vivo study | In mice bearing breast cancer xenografts, ABC294640 (100 mg/kg, p.o.) significantly reduced tumor growth in association with depletion of S1P levels. In severe combined immunodeficient mice bearing A- 498 xenografts, ABC294640 prolongs tumor growth and increases autophagy markers. ABC294640 prevents liver transplantation-induced inflammation and crosstalk between innate and acquired immunity, major substance precipitation and worsening of transplant injury, and improves liver function and survival rate. |
