introduction | 2-chloro -3, 6-difluorobenzaldehyde is a pharmaceutical intermediate, which can be obtained by reacting 2-chloro -1, 4-difluorobenzene with N,N-dimethylformamide after hydrogen extraction by lithium diisopropylamino. |
use | 2-chloro -3, 6-difluorobenzaldehyde is an aldehyde derivative, which can be used as a pharmaceutical intermediate to prepare downstream products 2-chloro -3, 6-difluorobenzonitrile. |
Preparation | Dissolve 2-chloro-1, 4-difluorobenzene (1.78g,12.0mmol) in tetrahydrofuran (50mL) and cool to -78°C. Lithium diisopropylamide (1.8M in tetrahydrofuran, 7.3mL,13.2mmol) was added. Stir at -78 ℃ for 30 minutes, add N,N-dimethylformamide (1.05g,14.4mmol). Stir at -78°C for a further 15 minutes, add acetic acid (3mL) and water (100mL) and heat the mixture to RT. After extraction with ethyl acetate, the organic phase was washed with 1M hydrochloric acid solution and brine and dried with magnesium sulfate. Vacuum concentration produces 2-chloro-3, 6-difluorobenzaldehyde (1.51g,71% theoretical value). |
