In vitro study | S1P acts on K562 cells and inhibits NK cell lysis. Various concentrations of FTY720 reversed the inhibitory effect of S1P with an IC50 of 173 nM. In addition, incubation at 10 nM FTY720 alone did not have any effect on the expression of costimulatory molecules. By comparing the effect of S1P with the combined effect of S1P and FTY720, FTY720 can reverse the increase in HLA-I expression induced by S1P, while the cell percentage and MFI are also improved. Medium and high dose FTY720-P also enhanced TGF-β1 levels. The expression of TGF-β1 and Foxp3 mRNA was up-regulated in the high dose FTY720-P group. Effector (Teff) cell proliferation was inhibited in medium and high dose FTY720-P groups with a Treg/Teff cell ratio of 1:1. At a ratio of 1:1, Teff cells in the high dose FTY720 group were also inhibited. |
