Introduction | (S)-1-BOC-pyrrolidine -3-formic acid, also known as (S)-N-Boc-β-proline, is an amino acid derivative, which can be prepared by one-step reaction of (S)-β-proline and Boc2O. There are reports that it can be used to prepare USP30 inhibitors. |
Preparation | Suspended (S)-β-proline (152mg,1.32mmol) and Boc2O(350mg,1.58mmol) on deionized H2O(1mL) and acetone (3mL). The mixture was cooled to 0°C and solid Na2CO3(72mg,0.660mmol) was added. After 3 hours, acetone was removed by vacuum and citric acid aqueous solution (20% w/v) was added dropwise until pH4. The mixture was diluted with H2O(15mL) and EtOAc(15mL). The aqueous phase was extracted with EtOAc(2 × 10mL). The combined organic phase was dried with Na2SO4 and concentrated, after which the product was precipitated into a white solid, giving 284mg(99%) of (S)-1-BOC-pyrrolidin-3-formic acid. |
